چکیده

Methods for the synthesis of bromodeoxyuridine-Br s2 and bromodeoxyuridine-H a (BUdR) are described. The incorporation of both thymidine and BUdR into the deoxyribonucleic acid (DNA) of rat bone marrow cells in vitro was markedly influenced by concentrations of thymidine and BUdR, respectively, in the incubation media. At equivalent concentrations of respective substrate, the amount of thymidine incorporated always exceeded that of BUdR. Limiting concentrations of both thymidine and BUdR were found, above which little or no further incorporation of the respective compounds occurred. Efficiency of incorporation of each compound, as measured by percentage of the available dose which was incorporated, was highest at low respective substrate concentrations. The rates of incorporation of thymidine and BUdR into the DNA of Ehrlich ascites tumor cells in vitro were maximal at the beginning of incubation and fell progressively thereafter. Thymidine and BUdR mutually inhibit the incorporation of the other into DNA. Evidence is presented that this inhibition may be due to competition by the compounds for binding sites on the same enzyme, which is rate-limiting for their entry into the cell and subsequent incorporation into DNA. The presence of 5-fluorouracil in the medium exerts an enhancing effect on thymidine and BUdR incorporation.

[Methyl-14C]-choline incorporation into nude mice bearing tumor xenografts-correlation with [methyl-3H]-thymidine: A pilot study

Background: Many in vitro studies suggested that choline incorporation into many types of tumors is related to cell proliferation. Whether in vivo choline incorporation is related also to cell proliferation or not was my question. Therefore, the aim of this pilot study was to investigate the relationship of in vivo [methyl- 14C]-choline incorporation and in vivo [methyl-3H]- thymidine incorpora...

Specificity and efficiency of thymidine incorporation in Escherichia coli lacking thymidine phosphorylase.

A mutant of Escherichia coli lacking the catabolic enzyme thymidine phosphorylase readily incorporates exogenous thymidine into deoxyribonucleic acid (DNA) even when provided at concentrations as low as 0.2 mug/ml. Incorporation by this prototrophic strain occurs specifically into DNA, since, with radioactively labeled thymidine, (i) more than 98% is incorporated into alkali-stable material, (i...

Introduction of the 5-halogenated uracil moiety into deoxyribonucleic acid of mammalian cells in culture.

Introduction of the 5-halogenated uracil moiety into the deoxyribonucleic acid of several bacterial strains has been described by Weygand and Wacker (l), Zamenhof and Griboff (2), and Dunn and Smith (3). A corresponding introduction into the deoxyribonucleic acid of mammalian cells was recently reported by Eidinoff et al. (4, 5, 6) and by Prusoff (7) when 5-bromodeoxyuridine or 5-iododeoxyuridi...

Studies on the biosynthesis of deoxyribonucleic acid by soluble mammalian enzymes.

A variety of reactions associated with the anabolism of ribonucleotides and deoxyribonucleotides as well as with their incorporation into ribonucleic acid and deoxyribonucleic acid, respectively, has been shown to occur in the high speed, particle free, supernatant fraction of rat liver. Such a cellular fraction obtained from normal rat liver has been shown to convert UMP (I), dAMP, dCMP, and d...

Selective Inhibition by Sorbylhydroxamic Acid of Deoxyribonucleic Acid Synthesis in Ehrlich Ascites Tumor Cells1

Sorbylhydroxamic acid (SOHA) inhibited the incorporation in vitro of thymidine into deoxyribonucleic acid in Ehrlich ascites tumor cells, with no appreciable effect on the incorporation of uridine into ribonucleic acid or of L-leucine into protein. Hexanoylhydroxamic acid, devoid of the conjugated double-bond system, had no such activity. The concentration of SOHA conferring 50% inhibition of D...
